== Overview == Smoking most commonly leads to diseases affecting the heart and lungs and will commonly affect areas such as hands or feet. First signs of smoking-related health issues often show up as numbness in the extremities, with smoking being a major risk factor for heart attacks, chronic obstructive pulmonary disease (COPD), emphysema, and cancer, particularly lung cancer, cancers of the larynx and mouth, and pancreatic cancer. The immune system is also weakened by smoking, which makes the body more susceptible to infections and takes longer to recover from injuries. Overall life expectancy is also reduced in long term smokers, with estimates ranging from 10 to 17.9 years fewer than non-smokers. About half of men who smoke long-term will die of illness due to smoking. The association of smoking with lung cancer and COPD is among the strongest, both in the public perception and etiologically. Among male smokers, the lifetime risk of developing lung cancer is 17%; among female smokers, the risk is 12%. This risk is significantly lower in non-smokers: 1.3% in men and 1.4% in women. For COPD, the 25 year incidence of moderate and severe COPD is at least 21% for continuous smokers and 4% for non-smokers, with no difference being reported between men and women. A person's increased risk of contracting disease is related to the length of time that a person continues to smoke as well as the amount smoked. However, even smoking one cigarette a day raises the risk of coronary heart disease by about 50% or more, and for stroke by about 30%.

Lithium fluoride had already been proposed in the USA in 1950 by Farrington Daniels (1889-1972), Charles A. Boyd and Donald F. Saunders (1924-2013) for solid-state dosimetry using thermoluminescent dosimeters. The intensity of the thermoluminescent light is proportional to the amount of radiation previously absorbed. This type of dosimetry has been used since 1953 in the treatment of cancer patients and wherever people are occupationally exposed to radiation. The thermoluminescence dosimeter was followed by OSL dosimetry, which is not based on heat but on optically stimulated luminescence and was developed by Zenobia Jacobs and Richard Roberts at the University of Wollongong (Australia). The detector emits the stored energy as light. The light output, measured with photomultipliers, is then a measure of the dose.
==== Peptidoglycan recognition proteins ==== PGLYRPs are conserved from insects to mammals. Mammals produce four secreted soluble peptidoglycan recognition proteins (PGLYRP-1, PGLYRP-2, PGLYRP-3 and PGLYRP-4) that recognize muramyl pentapeptide or tetrapeptide. They can also bind to LPS and other molecules by using binding sites outside of the peptidoglycan-binding groove. After recognition of peptidoglycan, PGLYRPs activate polyphenol oxidase (PPO) molecules, Toll, or immune deficiency (IMD) signalling pathways. That leads to production of antimicrobial peptides (AMPs). Each of the mammalian PGLYRPs display unique tissue expression patterns. PGLYRP-1 is mainly expressed in the granules of neutrophils and eosinophils. PGLYRP-3 and 4 are expressed by several tissues such as skin, sweat glands, eyes or the intestinal tract. PGLYRP-1, 3 and 4 form disulphide-linked homodimers and heterodimers essential for their bactericidal activity. Their binding to bacterial cell wall peptidoglycans can induce bacterial cell death by interaction with various bacterial transcriptional regulatory proteins. PGLYRPs are likely to assist in bacterial killing by cooperating with other PRRs to enhance recognition of bacteria by phagocytes. PGLYRP-2 is primarily expressed by the liver and secreted into the circulation. Also, its expression can be induced in skin keratinocytes, oral and intestinal epithelial cells. In contrast with the other PGLYRPs, PGLYRP-2 has no direct bactericidal activity.

These changes can account for the physical and psychological remission of the pathology of addiction. The longer effects of KOR agonism (30 minutes or greater) have been linked to KOR-dependent stress-induced potentiation and reinstatement of drug seeking. Implicating the KOR/dynorphin system as an anti-reward system, supported by the role of KOR signaling and stress, mediating both stress-induced potentiation of drug reward and stress-induced reinstatement of seeking behavior. This in turn addresses what was thought to be paradoxical above. That is, rather, KOR signaling is activated/upregulated by stress, drugs of abuse and agonist administration - resulting in negative affective state. As such drug addiction is maintained by avoidance of negative affective states in stress, craving, and drug withdrawal.
== Mechanism of action == Tigecycline is a broad-spectrum antibiotic that acts as a protein synthesis inhibitor. It exhibits bacteriostatic activity by binding to the 30S ribosomal subunit of bacteria and thereby blocking the interaction of aminoacyl-tRNA with the A site of the ribosome. In addition, tigecycline has demonstrated bactericidal activity against isolates of S. pneumoniae and L. pneumophila. Studies have shown that tigecycline binds to the 70S ribosome with 5 fold and >100 fold greater affinity than minocycline and tetracycline, respectively . As previously mentioned, tigecycline still binds to the A site of the 30S ribosomal subunit, however the binding of the novel antibiotic involves substantial interactions with residues of helix H34 of that same subunit. These interactions are not observed in the binding of tetracycline. The findings indicate that tigecycline likely has a unique mechanism of action that prevents inhibition from ribosomal protection. It is a third-generation tetracycline derivative within a class called glycylcyclines which carry a N,N-dimethyglycylamido (DMG) moiety attached to the 9-position of tetracycline ring D. With structural modifications as a 9-DMG derivative of minocycline, tigecycline has been found to improve minimal inhibitory concentrations against Gram-negative and Gram-positive organisms, when compared to tetracyclines.
=== Response to Russia and China === On March 6, 2026, Hegseth dismissed reports from CBS News and other outlets citing U.S. officials who claimed Russia was providing intelligence to Iran regarding American positions in the Middle East. Hegseth stated, "We're putting the other guys in danger, and that's our job. So we're not concerned about that. ... But the only ones that need to be worried right now are Iranians that think they're gonna live." However, four days later, he warned Russia against intervention following further reports of its alleged assistance in targeting U.S. military assets in the Middle East. While Hegseth initially dismissed China as "irrelevant" to the conflict in early March 2026, his stance shifted by the end of the month following reports of alleged technological support to Tehran. On March 31, he stated the U.S. was "addressing and mitigating" Chinese involvement. However, on April 16, Hegseth announced that Beijing had provided high-level assurances to the White House that it would not send weapons to Iran, citing direct communication between President Trump and Xi Jinping ahead of a planned summit in Beijing.
