Semaglutide is a synthetic peptide analogue of glucagon-like peptide-1, a gut hormone released after nutrient intake. The molecule contains 31 amino acid residues and differs from the native sequence at several positions. A non-natural residue at position eight resists the enzyme that normally truncates the hormone, while a lysine-linked fatty diacid side chain promotes binding to serum albumin. These two modifications extend the circulating half-life from minutes to roughly one week. The peptide is produced by solid-phase synthesis followed by selective acylation, and its identity and purity are confirmed by spectrometric and chromatographic techniques.
The primary target is the GLP-1 receptor, a class B G protein-coupled receptor expressed on pancreatic beta cells, in the gut, and in several brain regions. Receptor activation raises intracellular cyclic AMP, which potentiates glucose-dependent insulin secretion and lowers glucagon release when blood glucose is elevated. Signalling in the hypothalamus and brainstem is associated with reduced appetite and slower gastric emptying. Because the insulinotropic effect depends on prevailing glucose levels, the hypoglycaemic risk of the peptide alone is described as low in most study settings. The relative contribution of peripheral and central actions remains an active research question.
Storage at minus 20 degrees Celsius or lower in a desiccated container preserves the peptide for extended periods, while working solutions are commonly held at two to eight degrees Celsius for short intervals. Light exposure and repeated freeze-thaw cycles accelerate degradation, so dividing material into single-use aliquots is generally recommended. Adsorption to glass and plastic surfaces can lower the measured concentration of dilute solutions, particularly below one milligram per millilitre. The degradation routes most often reported for GLP-1 analogues are deamidation, methionine oxidation, and backbone hydrolysis. Relative rates under specific conditions are frequently described only for individual formulations.
Reverse-phase high-performance liquid chromatography with ultraviolet detection near 214 or 280 nanometres is widely used to assess purity and to resolve related impurities. Liquid chromatography coupled to mass spectrometry confirms identity through the protonated molecular ion and fragment ions formed in tandem experiments. Capillary electrophoresis and peptide mapping after enzymatic digestion supply complementary information on charge variants and modification sites. Circular dichroism and nuclear magnetic resonance can report on secondary structure in solution. Absolute quantification usually depends on an external standard, and reported purity depends on the detection wavelength and integration parameters chosen.

Lyophilized semaglutide is typically stored at temperatures between minus 20 and minus 80 degrees Celsius for long-term preservation. Short-term storage at 2 to 8 degrees Celsius is common for working aliquots. Repeated freeze-thaw cycles can degrade the peptide and are usually avoided. The molecule is hygroscopic in its solid form, so containers should remain sealed with desiccant. Solutions are less stable than powders and are generally prepared fresh. Light exposure is limited because aromatic residues can undergo photo-oxidation.
Semaglutide dissolves readily in water and in aqueous buffers near neutral pH. Solubility decreases near the isoelectric point, where net charge is minimal. Common laboratory solvents include phosphate-buffered saline and dilute ammonium bicarbonate. Strongly acidic or basic conditions may accelerate hydrolysis. Working concentrations are usually prepared by diluting a concentrated stock. Vial surfaces can adsorb small amounts of peptide at low concentrations, so carrier proteins or low-binding tubes are sometimes used.
Reverse-phase high-performance liquid chromatography is the standard method for purity assessment, separating the peptide from truncated or oxidized variants. Mass spectrometry confirms molecular mass and detects modifications, while ultraviolet absorbance near 280 nanometers supports concentration measurement through tryptophan and tyrosine residues. Circular dichroism can indicate secondary structure, though the peptide is largely helical in solution, and ion-exchange chromatography resolves charge variants. Purity values above 95 percent are typical for research-grade material. Stability studies track degradation over time under defined conditions.
Semaglutide is a synthetic peptide that acts as an agonist at the glucagon-like peptide-1 receptor. It is a structural analogue of human GLP-1(7-37), modified to resist enzymatic degradation by dipeptidyl peptidase-4. The peptide backbone contains alpha-aminoisobutyric acid at position 8, a substitution that stabilises the helix and slows cleavage. A fatty diacid side chain attached through a linker at lysine 34 promotes binding to serum albumin, which extends the circulating half-life. These two modifications together allow less frequent administration than native GLP-1 requires.
Activation of the GLP-1 receptor couples to Gs signalling and raises intracellular cyclic AMP in pancreatic beta cells. The resulting insulin release depends on prevailing glucose concentrations, so the effect is greater when glucose is elevated. Receptor engagement also suppresses glucagon secretion and slows gastric emptying, which flattens post-meal glucose excursions. In the central nervous system, signalling in hypothalamic and brainstem regions is associated with reduced appetite and lower energy intake. Studies continue to examine effects on cardiac, renal and hepatic endpoints; whether those benefits are independent of weight change remains an open question.
Clinical development of this compound followed earlier short-acting GLP-1 analogues that required frequent injection. Once-weekly subcutaneous formulations entered use after 2017, and an oral formulation using a permeation enhancer later became available. The oral version pairs the peptide with sodium N-(8-[2-hydroxybenzoyl] amino) caprylate, a carrier that improves uptake across the gastric epithelium. Interest has expanded from glycaemic control into weight management and metabolic liver disease. Regulatory status and approved indications differ between countries, and the product remains subject to ongoing safety monitoring.
===== Oceania ===== New Zealand - DMT is classified as a Class A drug under the Misuse of Drugs Act 1975. Australia - DMT is listed as a Schedule 9 prohibited substance in Australia under the Poisons Standard (October 2015). A Schedule 9 drug is outlined in the Poisons Act 1964 as "Substances which may be abused or misused, the manufacture, possession, sale, or use of which should be prohibited by law except when required for medical or scientific research, or for analytical, teaching or training purposes with approval of the CEO". Between 2011 and 2012, the Australian federal government was considering changes to the Australian Criminal Code that would classify any plants containing any amount of DMT as "controlled plants". DMT itself was already controlled under current laws. The proposed changes included other similar blanket bans for other substances, such as a ban on any and all plants containing mescaline or ephedrine. The proposal was not pursued after political embarrassment on realisation that this would make the official floral emblem of Australia, Acacia pycnantha (golden wattle), illegal. The Therapeutic Goods Administration and federal authority had considered a motion to ban the same, but this was withdrawn in May 2012 (as DMT may still hold potential entheogenic value to native and/or religious people). Under the Misuse of Drugs Act 1981 6.0g (3/16oz) of DMT is considered enough to determine a court of trial and 2.0g (1/16oz) is considered intent to sell and supply. In December 2004, the U.S.
Inhibition of radical SAM enzyme MqnE in menaquinone biosynthesis is reported to be an effective antibacterial strategy against H. pylori. Radical SAM enzyme BlsE has been discovered to be a central enzyme in blasticidin S biosynthetic pathway. Blasticidin S produced by Streptomyces griseochromogenes exhibits strong inhibitory activity against rice blast caused by Pyricularia oryzae Cavara. This compound specifically inhibits protein synthesis in both prokaryotes and eukaryotes through inhibition of peptide bond formation in the ribosome machinery. A new fungal radical SAM enzyme has also been reported to facilitate the biocatalytic routes for synthesis of 3'-deoxy nucleotides/nucleosides. 3'deoxynucleotides are a class of drugs that interfere with the metabolism of nucleotides, and their incorporation into DNA or RNA terminates cell division and replication. This activity explains why this compound is an essential group of antiviral, antibacterial or anticancer drug.
=== Development === The series originated from discussions between Toho producers and Yeon Sang-ho in 2018, during which Yeon selected The Human Vapor from Toho's "Transforming Human Series" for its blend of speculative science and human drama. Yeon, known for global hits like Train to Busan (2016) and Netflix's Hellbound (2021), emphasized the original film's "skillful sci-fi expressions" and emotional core, stating: "While it is a sci-fi thriller, at its core, it is a story about people. We focused on treating human emotions with care and portraying the characters’ humanity." Script development took three years, with Ryu Yong-jae—Yeon's collaborator on Parasyte: The Grey (2024)—joining as co-writer. Shinzo Katayama, who was acclaimed for his directing live action TV seriesGannibal (2022), was selected as director for his ability to merge genre elements with intimate storytelling. Katayama, upon revisiting the original, noted: "I was intrigued by its blend of human drama and romantic elements, despite featuring an absurd creature like the Human Vapor." The project represents Netflix's first partnership with Toho, aiming to revive the studio's tokusatsu legacy for global audiences using cutting-edge VFX. Wow Point, the production company behind Parasyte: The Grey, co-produces, infusing Korean expertise in genre storytelling.
=== Enzyme production === A significant milestone in the use of K. phaffii in food technology was its GRAS classification by the US Food and Drug Administration, alongside their approval of recombinant proteins. It is now used to produce various enzymes that serve as processing aids and food additives. In bakery production, enzymes produced by genetically modified Komagataella can maintain bread freshness and compensate for variations in flour and malt quality. In brewing, they can reduce beer's alcohol content or modify the flavour and filtration properties of wine. Recombinant expressed phospholipase C is used to degum high-phosphorus vegetable oils by hydrolysing phospholipids. In animal feed, phytase produced by K. phaffii breaks down phytic acid, an antinutrient. Recently, K. phaffii has been used increasingly to produce soy leghemoglobin, a plant heme protein that gives plant-based meat analogues their colour and flavour. This expands the industrial application of K. phaffii from technical enzymes towards functional food ingredients. For this purpose, the yeast utilises its strong methanol-inducible promoters and efficient secretion to produce functional leghemoglobin for use as a food ingredient. Through strain engineering, including optimisation of heme biosynthesis and signal peptides, and controlled fed-batch fermentation, gram-per-litre titres of secreted leghemoglobin have been achieved. This makes large-scale production for meat analogues economically viable.

Anxiety disorders Bobble-head doll syndrome – a sensation that the skull may fall off the cervical spine Clumsiness and motor delay Cognitive and memory decline Double or blurred vision Dysphagia – a sensation of choking Dyspnea Fatigue Lhermitte's sign Migraine Nausea Neck, shoulder, and jaw pain Occipital headaches Orthostatic intolerance Photophobia Syncope Tenderness at base of skull Tinnitus Tremors Palpitations Vertigo or dizziness Weakness of limbs Symptoms are frequently worsened by a Valsalva maneuver or by being upright for long periods of time. Being upright is problematic because gravity allows increased interaction between the brain stem and the top of the spinal column, increasing symptoms. Lying in the supine position can bring short-term relief. Lying supine eliminates the downward gravitational pull, reducing symptoms to some degree. Lying with the feet somewhat higher and head lower can be helpful in symptom reduction.

Rhodesian politicians frequently reminded officials in the other two nations of common security interests based on the similarity of their restive internal situations. They saw strong parallels between their nation's position of being threatened by black nationalist insurgencies and the Portuguese predicament with FRELIMO in Mozambique, as well as to a lesser extent the insurgencies in South Africa and South West Africa. Under the auspices of the Alcora Exercise, the three countries' bureaucracies began routinely sharing information and seeking common diplomatic positions. Lieutenant General Alan Fraser, a senior strategist in the South African Defence Force wrote in 1970, "there can be no doubt in any of our minds that we have a common enemy: we, i.e. Portugal, the RSA and Rhodesia. Unless we are to lay ourselves open to the possibility of defeat in detail, we must fight this enemy jointly—if not simultaneously." Nevertheless, aside from intelligence-sharing and some limited coordination on the operational level in Mozambique, the Portuguese could offer Rhodesia little decisive assistance. Portuguese colonial administrators in Mozambique were preoccupied with FRELIMO and somewhat depleted by a decade of war, and little could be spared to assist a foreign ally. Rhodesia expected far more from South Africa, which possessed greater military resources and diplomatic influence abroad at the time. After the Carnation Revolution and the end of Portuguese rule in Mozambique in 1975, it was no longer viable for the Smith regime to sustain white minority rule indefinitely.
The spread of ironworking in Central and Western Europe is associated with Celtic expansion. According to Pliny the Elder, iron use was common in the Roman era. In the lands of what is now considered China, iron appears approximately 700–500 BC. Iron smelting may have been introduced into China through Central Asia. The earliest evidence of the use of a blast furnace in China dates to the 1st century AD, and cupola furnaces were used as early as the Warring States period (403–221 BC). Usage of the blast and cupola furnace remained widespread during the Tang and Song dynasties. During the Industrial Revolution in Britain, Henry Cort began refining iron from pig iron to wrought iron (or bar iron) using innovative production systems. In 1783 he patented the puddling process for refining iron ore. It was later improved by others, including Joseph Hall.
The Special Program of Assisted Reproduction (SPAR) is a program offered to HIV discordant couples (serodiscordant) at the Bedford Research Foundation's clinical laboratory. The program takes advantage of ART (assisted reproduction technology) procedures (including "sperm washing") to assist couples achieve a pregnancy who would otherwise risk transmitting the father's HIV infection to the mother and the child through intercourse. SPAR employs extremely stringent safety standards; samples will only be submitted for "sperm washing" and cryopreservation that have first been screened for HIV virus particles and infected cells by specialized molecular biology tests. Only samples that have been determined to have an undetectable HIV viral burden will be submitted for infertility procedures. Assisted reproductive technologies (ART), such as in vitro fertilization (IVF), treat many disorders of the female, but until recent years, treatment options for male factor infertility were limited, and donor sperm was routinely recommended to achieve a pregnancy. Advances in ART, however, have created new possibilities for men with male factor infertility, including those whose sperm counts have been decreased by cancer treatment and other diseases, and men with incurable, sexually transmissible virus diseases. Infectious diseases transmissible by semen for which there is no cure include:

Native GLP-1 is degraded within minutes by dipeptidyl peptidase-4 and neutral endopeptidases. Semaglutide carries a non-natural amino acid at position eight that blocks that cleavage, and a fatty diacid side chain that binds albumin. The result is a much longer duration of action than the native hormone.
The fatty diacid chain associates strongly with serum albumin, which keeps the peptide in circulation and shields it from rapid renal clearance. Bound peptide is released gradually, producing a sustained receptor signal. This mechanism also reduces the peak-to-trough variation seen with shorter-acting analogues.
The balance between peripheral receptor activation and signalling in the central nervous system is not fully resolved. The extent to which slowed gastric emptying accounts for reduced energy intake, compared with direct effects on appetite circuits, is debated. Long-term effects on lean mass and on tissues outside the gut and brain are still under study.
Filtering removes particulate matter that can block columns or scatter light. A 0.22 micrometre membrane is typical, and the filter material should be checked for peptide adsorption.
