They use DNA template encoding the gene of interest fused with GST protein, and it was immobilized in the solid surface. Anti-GST antibody and biotinylated plasmid DNA were bounded in aminopropyltriethoxysilane (APTES)-coated slide. BSA can improve the binding efficiency of DNA. Biotinylated plasmid DNA was bound by avidin. New protein was synthesized by using cell-free expression system i.e. rabbit reticulocyte lysate (RRL), and then the new protein was captured through anti-GST antibody bounded on the slide. To test protein–protein interaction, the targeted protein cDNA and query protein cDNA were immobilized in a same coated slide. By using in vitro transcription and translation system, targeted and query protein was synthesized by the same extract. The targeted protein was bound to array by antibody coated in the slide and query protein was used to probe the array. The query protein was tagged with hemagglutinin (HA) epitope. Thus, the interaction between the two proteins was visualized with the antibody against HA.
== Partitioning methods == The most common method of measuring amino acid hydrophobicity is partitioning between two immiscible liquid phases. Different organic solvents are most widely used to mimic the protein interior. However, organic solvents are slightly miscible with water and the characteristics of both phases change making it difficult to obtain pure hydrophobicity scale. Nozaki and Tanford proposed the first major hydrophobicity scale for nine amino acids. Ethanol and dioxane are used as the organic solvents and the free energy of transfer of each amino acid was calculated. Non liquid phases can also be used with partitioning methods such as micellar phases and vapor phases. Two scales have been developed using micellar phases. Vapor phases represent the simplest non polar phases, because it has no interaction with the solute. == Life == Richard Laurence Millington Synge was born in West Kirby on 28 October 1914, the son of Lawrence Millington Synge, a Liverpool stock-broker, and his wife, Katherine C. Swan. Synge was educated at the Old Hall in Wellington, Shropshire and at Winchester College. He then studied Chemistry at Trinity College, Cambridge. He spent his entire career in research, at the Wool Industries Research Association, Leeds (1941–1943), Lister Institute for Preventive Medicine, London (1943–1948), Rowett Research Institute, Aberdeen (1948–1967), and Food Research Institute, Norwich (1967–1976). It was during his time in Leeds that he worked with Archer Martin, developing partition chromatography, a technique used in the separation mixtures of similar chemicals, that revolutionised analytical chemistry. === Knoxville in the 1960s === In 1960, several Knoxville College students, led by Robert Booker and Avon Rollins, engaged in a series of sit-ins to protest segregation at lunch counters in Downtown Knoxville. This action prompted downtown department stores to desegregate, and by the end of the decade, most other downtown businesses had followed suit. City schools also gradually desegregated during this period, largely in response to a lawsuit brought by Josephine Goss in 1959. Between 1945 and 1975, the University of Tennessee's student body grew from just under 3,000 to nearly 30,000. The school's campus expanded to cover the entire area between Cumberland Avenue and the river west of Second Creek, and the Fort Sanders neighborhood was largely converted into student housing. By the mid-1970s, U.T. employed over 4,000 faculty and staff, providing a boost to the city's economy. The growing popularity of the school's sports teams led to the expansion of Neyland Stadium, one of the largest non-racing stadiums in the nation, and the eventual construction of Thompson–Boling Arena, one of the largest basketball venues in the nation at the time of its completion. While unemployment declined to just 2.8% in the 1960s, many of the jobs paid low wages, stunting the growth of the city's service sector.
