reversed-phase HPLC raises a handful of sensible questions. This page answers them in order, starting with the fundamentals and moving to applications.

Peptides are sensitive to temperature, light, oxygen, and repeated freeze-thaw cycles. Semaglutide in dry form is generally held at refrigerated temperatures, while reconstituted solutions require a defined short-term storage window. Vials should be kept in secondary packaging to limit photodegradation, and exposure to alkaline conditions is avoided because it accelerates chemical degradation. Adsorption to glass and some plastics can reduce the measured concentration of dilute solutions, so low-binding polypropylene containers are preferred for analytical work. Each transfer step introduces a small risk of contamination, and closed handling practices reduce that risk.
Routine characterisation of the peptide relies on reversed-phase high-performance liquid chromatography, often paired with ultraviolet detection near 214 nanometres. Related substances such as deamidated, oxidised, and truncated sequences elute at characteristic positions and are quantified by area percentage. Electrospray ionisation mass spectrometry confirms the molecular mass and can resolve some closely related variants. Peptide mapping after enzymatic digestion provides sequence-level verification and is useful when a full identity profile is required. Method parameters such as column chemistry, gradient, and mobile-phase pH influence the separation and must be reported alongside results.

As a peptide, semaglutide is sensitive to conditions that break amide bonds or modify side chains. Deamidation of asparagine and glutamine residues, oxidation of methionine and tryptophan, and non-covalent aggregation are the main degradation routes described in published stability work. Rate depends strongly on pH, buffer species, ionic strength, temperature and exposure to light. Formulators therefore choose a defined solution pH and often add excipients such as phosphate buffer, propylene glycol and phenol, each of which plays a separate role in pH control, tonicity or preservation.
Storage guidance for the finished injectable product distinguishes the unused state from the in-use state. Before first use, pens are kept refrigerated between 2 and 8 degrees Celsius, protected from light, and never frozen, since freezing can disrupt the peptide or the device. After first use, label instructions in several markets permit storage at room temperature up to about 30 degrees Celsius for a limited number of days. Solid research-grade material is normally held at or below minus 20 degrees Celsius, often with desiccant, and allowed to equilibrate before opening.
Identity and purity are usually assessed by reverse-phase high-performance liquid chromatography coupled to mass spectrometry. Retention time and observed mass are compared against a reference standard run under identical conditions. Impurity profiles reveal deamidation products, oxidized methionine variants, and truncated fragments that arise during synthesis or storage. Peptide mapping through enzymatic digestion confirms the primary sequence, while amino acid analysis offers an independent check on overall composition.
Stability studies examine how temperature, pH, and moisture influence degradation rates over time. In aqueous solution, hydrolysis and deamidation accelerate as pH moves away from mildly acidic conditions. Light exposure and residual metal ions can also trigger oxidation of susceptible residues. Accelerated aging at elevated temperature is used to estimate shelf life, though extrapolation to room temperature carries uncertainty because individual degradation pathways do not always scale predictably.
Degradation proceeds along several parallel routes. Deamidation of asparagine and glutamine residues generates charged variants that shift retention time in chromatographic analysis. Oxidation targets methionine and can be accelerated by trace metals or dissolved oxygen. Non-covalent aggregation produces dimers, oligomers, and larger species that are difficult to reverse. Isomerisation at aspartate residues is slower but measurable under thermal stress. The distribution among these pathways depends on pH, buffer composition, ionic strength, and the presence of excipients such as sugars or surfactants.
Reverse-phase high-performance liquid chromatography with ultraviolet detection near 214 nm is the standard purity method, reported as area percent. Mass spectrometry, usually with electrospray ionisation, confirms identity and reveals covalent modifications. Size-exclusion chromatography quantifies aggregates and fragments. Peptide mapping after enzymatic digestion localises changes to specific sequence regions. Circular dichroism and infrared spectroscopy report on secondary structure, while light scattering tracks particle formation in liquid formulations. No single technique captures every quality attribute.
Quality control relies on pharmacopoeial monographs where they exist, combined with in-house specifications for identity, purity, water content, and counter-ion composition. Reference standards allow calibration across laboratories, although certified materials for every analogue are not universally obtainable. Batch records, chromatograms, and mass spectra form the documentation trail. Regulatory classification varies by jurisdiction and intended use, and research-grade material differs from pharmaceutical-grade material in testing scope. Analytical uncertainty is often expressed as relative standard deviation across replicate injections.
The sequence incorporates alpha-aminoisobutyric acid at position 8, replacing the alanine found in the natural hormone. This substitution blocks the primary DPP-4 recognition site and contributes most of the enzymatic stability. Albumin binding further protects the peptide and reduces the frequency of administration required to maintain active plasma levels. Because the fatty acid chain increases lipophilicity, the compound is formulated as a solution rather than a simple aqueous buffer. Researchers describe the design as an incremental optimization of earlier GLP-1 analogs rather than a wholly new scaffold.
Reported molecular weight is approximately 4113.6 daltons for the free base, and the peptide is supplied as a lyophilized powder or in buffered liquid form depending on the intended use. It is freely soluble in water when formulated with appropriate excipients, though the unconjugated peptide shows limited stability at neutral pH over long periods. Analytical characterization typically relies on reversed-phase high-performance liquid chromatography and mass spectrometry. Purity specifications for research-grade material commonly exceed ninety-five percent by area. Isotopic and impurity profiles differ between suppliers.
=== Prenatal diagnosis === Mutations in the main genes responsible for TCS can be detected with chorionic villus sampling or amniocentesis. Rare mutations may not be detected by these methods. Ultrasonography can be used to detect craniofacial abnormalities later in pregnancy, but may not detect milder cases.
When cooling outdoor air, a cooling unit must deal with the air's sensible heat and latent heat. Typical vapor-compression air-conditioning (VCAC) units manage the latent heat in air through cooling fins held below the dew point temperature of the moist air at the intake. These fins condense the water, dehydrating and thus substantially reducing the air's heat content. Energy usage is dependent on the cooling coil's temperature and improves as the temperature of the coil rises above the dew point. This makes it desirable to handle dehumidification through means other than condensation. One such means is by adsorbing the water from the air into a desiccant coated onto the heat exchangers, using the waste heat exhausted from the unit to desorb the water from the sorbent and thus regenerate the desiccant. This is accomplished with two condenser/evaporator units through which the flow of refrigerant can be reversed once the desiccant is saturated, thus making the condenser the evaporator and vice versa. MOFs' high surface areas and porosities have made them the subject of research in water adsorption applications. Chemistry can help tune the optimal relative humidity for adsorption/desorption, and the sharpness of the water uptake. MOF CAU-10-H is reported to triple the performance of silica gel, the standard desiccant. CAU-10-H is reported to capture water at room temperature at relative humidity >18%. Heating the material to around 70 °C (158 °F) is sufficient to release the moisture, low enough to reach using solar heat or waste heat.
Cuban Masonic folklore once held that on March 27, 1818, at the Convent of San Francisco, there was a merger of the Temple of the Theological Virtues No. 1, Delicias, and Constancia to form the Spanish Grand Lodge of the York Rite (Spanish: Gran Logia Española del Rito York). However, most historians reject that this Grand Lodge was created in 1818. The more commonly accepted date for the creation of this Grand Lodge was in 1822. The secret government agent for the United States, Joel Roberts Poinsett, was a high-ranking a prominent member of the York Rite. Through him, the York Rite took hold in Mexico. Despite the fact that his private correspondences do not specifically reference Masonry in Cuba, he is still credited with developing the York Rite on the island, and the timeline of its development matches with his time spent here on fact-finding missions. In a letter dated March 5, 1822, there were 30 established York Right Lodges in Cuba; at least seven of them belonged to the Grand Lodge of Pennsylvania, two belonged to the Grand Lodge of South Carolina, and three belonged to the Grand Lodge of Louisiana. By 1822, the Blue Lodges under the Scottish Rite in Cuba were disgruntled with the lack of autonomy they held under the authority of the National Grand Orient of Spain, and believed that the York Rite granted more autonomy to their Lodges in Cuba. These Lodges entered into negotiations with the Grand Lodge of York Rite Masons, and began the process of merging.

=== Biosynthesis === The biosynthesis of the rapamycin core is accomplished by a type I polyketide synthase (PKS) in conjunction with a nonribosomal peptide synthetase (NRPS). The domains responsible for the biosynthesis of the linear polyketide of rapamycin are organized into three multienzymes, RapA, RapB, and RapC, which contain a total of 14 modules (figure 1). The three multienzymes are organized such that the first four modules of polyketide chain elongation are in RapA, the following six modules for continued elongation are in RapB, and the final four modules to complete the biosynthesis of the linear polyketide are in RapC. Then, the linear polyketide is modified by the NRPS, RapP, which attaches L-pipecolate to the terminal end of the polyketide, and then cyclizes the molecule, yielding the unbound product, prerapamycin.
=== Vincent of Beauvais, Albertus Magnus, and pseudo-Geber === Sulfuric acid was called 'oil of vitriol' by medieval European alchemists because it was prepared by roasting iron(II) sulfate or green vitriol in an iron retort. The first allusions to it in works that are European in origin appear in the thirteenth century AD, as for example in the works of Vincent of Beauvais, in the Compositum de Compositis ascribed to Albertus Magnus, and in pseudo-Geber's Summa perfectionis.
== Nomenclature == Various names have been applied to the undeclared conflict waged by South Africa in Angola and Namibia (then South West Africa) from the mid 1960s to the late 1980s. The term "South African Border War" has typically denoted the military campaign launched by the People's Liberation Army of Namibia (PLAN), which took the form of sabotage and rural insurgency, as well as the external raids launched by South African troops on suspected PLAN bases inside Angola or Zambia. These occasionally provoked conventional engagements with the Zambian National Defence Force (ZNDF) and the People's Armed Forces of Liberation of Angola (FAPLA). The strategic situation was further complicated by the fact that South Africa occupied large swathes of Angola for extended periods in support of the National Union for the Total Independence of Angola (UNITA), making the "Border War" an increasingly inseparable conflict from the parallel Angolan Civil War. "Border War" entered public discourse in South Africa during the late 1970s; the term was adopted thereafter by the country's ruling National Party. Due to the covert nature of most South African Defence Force (SADF) operations inside Angola, the term was favoured as a means of omitting any reference to engagements on foreign soil. Where tactical aspects of various engagements were discussed, some contemporary historians simply identified the conflict as the "bush war".

== Destruction and decay == 236U, on absorption of a thermal neutron, does not fission, but becomes 237U, which quickly beta decays to 237Np. However, the neutron capture cross section of 236U is low, and this process does not happen quickly in a thermal reactor. Spent nuclear fuel typically contains about 0.4% 236U. With a much greater cross-section, 237Np may eventually absorb another neutron and become 238Np, which quickly beta decays to plutonium-238 (another fissile isotope). 236U and most other actinide isotopes are fissionable by fast neutrons in a nuclear bomb or a fast neutron reactor. A small number of fast reactors have been in research use for decades, but widespread use for power production is still in the future. Uranium-236 alpha decays with a half-life of 23.42 million years to thorium-232. It is longer-lived than any other artificial actinides or fission products produced in the nuclear fuel cycle. (Plutonium-244, which has a half-life of 81.3 million years, is not produced in significant quantity by the nuclear fuel cycle, and the longer-lived uranium-235, uranium-238, and thorium-232 occur in nature.)
== Clinical significance == Furin is one of the proteases responsible for the proteolytic cleavage of HIV envelope polyprotein precursor gp160 to gp120 and gp41 prior to viral assembly. This protease is also thought to play a role in tumor progression. The use of alternate polyadenylation sites has been found for the FURIN gene. Furin is enriched in the Golgi apparatus, where it functions to cleave other proteins into their mature/active forms. Furin cleaves proteins just downstream of a basic amino acid target sequence (canonically, Arg-X-(Arg/Lys) -Arg'). In addition to processing cellular precursor proteins, furin is also used by a number of pathogens. For example, the envelope proteins of viruses such as HIV, influenza, dengue fever, several filoviruses including ebola and marburg virus, and the spike protein of SARS-CoV-2, must be cleaved by furin or furin-like proteases to become fully functional. When SARS-CoV-2 virus is being synthesized in an infected cell, furin or furin-like proteases cleave the spike protein into two portions (S1 and S2), which remain associated. Anthrax toxin, Pseudomonas exotoxin, and papillomaviruses must be processed by furin during their initial entry into host cells. Inhibitors of furin are under consideration as therapeutic agents for treating anthrax infection. Furin is regulated by cholesterol and substrate presentation. When cholesterol is high, furin traffics to GM1 lipid rafts. When cholesterol is low, furin traffics to the disordered region. This is speculated to contribute to cholesterol and age dependent priming of SARS-CoV.
The death of Yazid in 683 led to a major leadership crisis in the caliphate, and "the power of his house seemed to collapse everywhere", in the words of Orientalist Julius Wellhausen. Ubayd Allah initially neglected to support Yazid's son and designated successor, Mu'awiya II and secured oaths of allegiance to himself from the Basran Arab nobility. In a speech addressed to them, he emphasized his connection to Basra and promised to maintain the wealth of the city's inhabitants. Nonetheless, the Basrans turned against him, forcing him to abandon his palace. He was replaced by Abd Allah ibn al-Harith, a member of the Banu Hashim. Ubayd Allah took refuge with the Azdi chieftain Mas'ud ibn Amr in late 683 or early 684. He plotted to restore his governorship by encouraging Mas'ud to form an alliance of the Yamani and Rabi'a tribes against his opponents from the Banu Tamim and Ibn al-Harith. Mas'ud took to the pulpit of Basra's mosque to stir up the revolt, but Tamimi tribesmen under Ibn al-Harith and their asawira allies under Mah-Afridhun, stormed the building and killed Mas'ud. After Mas'ud's death, Ubayd Allah fled the city practically alone in March 684, taking the Syrian desert route to Hawran or Palmyra. In his rush to escape, he left his wife and family behind. When Ubayd Allah arrived in Syria, he found it in political disarray; Caliph Mu'awiya II had died weeks into his rule and a power vacuum ensued with many Syrian noblemen, particularly from the Qaysi tribes, switching allegiance to the rival, Mecca-based caliphate of Abd Allah ibn al-Zubayr.

Different techniques detect different classes of impurities, so a single number does not describe a sample completely. Reversed-phase chromatography resolves related peptides well but can miss inorganic salts, while mass spectrometry confirms mass without quantifying everything present. Comparing results requires knowing which method was used and how it was validated.
Cycling between frozen and liquid states can promote aggregation and surface adsorption at the container wall. Each cycle exposes the peptide to transient concentration and pH shifts near the ice interface. Aliquoting before storage limits the number of cycles a single container experiences.
Purity describes how much of the material is the intended substance, while identity describes whether that substance is the correct molecule. Mass spectrometry gives an observed mass that is compared with the theoretical value for the sequence. Peptide mapping after digestion adds sequence-level confirmation that mass alone cannot provide.
